VARIAÇÃO SAZONAL E POTENCIAL ANTICÂNCER DE
TIOFENOS ISOLADOS DE DUAS ESPÉCIES DE Tagetes: T. patula L. e T. erecta L.

Tiofenos são compostos comuns encontrados em Tagetes, eles são caracterizados pela presença de um ou mais anéis heterocíclicos contendo enxofre. Este trabalhou teve como objetivo quantificar a produção de tiofenos durante o desenvolvimento da Tagetes patula L. e em cada parte diferente da planta (folhas, flores, raízes e caúle) e isolar os tiofenos da espécie Tagetes erecta L. e avaliar a sua atividade citotóxica contra diferentes linhagens celulares de câncer. Os compostos foram identificados por Cromatografia gasosa acoplada a espectroscopia de massas. Os tiofenos foram identificados como 5-(4-hidroxi-1-butinil) 2,2’bitienil (BBTOH), 5-(4-acetoxi-1-butinil)-2-2’-bitienil BBTOAc, 5-(3-buten-1-enil)- 2,2’-bitienil (BBT), 2,2’:5’,2”-tertiofeno (α-T) e T-5. BBTOH, BBTAOc e α-T foram ativos e estatisticamente semelhantes contra as células de câncer de ovário (OVCAR8) e a fração tiofênica foi estatisticamente menos ativa. Em relação a linhagem celular do câncer do cólon e o câncer do sistema nervoso central, todas as drogas testadas mostraram ser citotóxicas e estatisticamente semelhantes. O padrão doxorubicina foi estatisticamente mais ativa que todos os compostos testados. Esses resultados são muito significativos, pois eles estão abaixo dos valores esperados pelo Instituto Nacional do Câncer dos Estados Unidos para ser uma boa droga antineoplásica. Os tiofenos causaram hemólise somente nas concentrações acima de 250 Og/mL. Em relação a quantificação durante o desenvolvimento da planta, pode ser concluído que a maior concentração média é alcançada no dia 60 e a partir desse dia a concentração de tiofenos diminui. Em relação as diferentes partes da planta, a parte com maior concentração de tiofenos são as Raízes, seguido pelo caule, flores e folhas. A concentração de cada tiofenos varia muito em relação a cada parte da planta. Então, pode ser concluído que a concentração de tiofenos varia durante o desenvolvimento da planta, no entanto, a concentração atinge um pico por volta do dia 60 e após a concentração diminui. Em relação a citotoxicidade, os tiofenos da Tagetes erecta mostraram boa toxicidade em relação as linhagens tumorais e uma baixa toxicidade contra os eritrócitos. No geral, todos os tiofenos isolados exibiram atividades consideradas significantes de acordo com o NCI, mostrando que os tiofenos da Tagetes erecta representam uma importante classe de drogas antineoplásicas. Abstract:
Thiophenes are common compounds present in Tagetes, characterized by the presence of one or more sulfur-containing heterocyclic rings. This work aimed to quantify the thiophene production during the development of Tagetes Patula L. and in each part of the plant (leaves, flowers, roots and stem) and to isolate thiophenes from Tagetes erecta L. and evaluate the cytotoxic activity against some cancer cell types. The compounds were identified by Gas Chromatography coupled with Mass Spectrometry. The thiophenes were identified as 5-(4-hydroxy-1-butynyl) 2,2’bityenyl (BBTOH), 5-(4-acetoxy-1-butynyl)-2- 2’-bityenil (BBTOAc) and 2,2’:5’,2”-terthiophene (α-T). 5-(3-buten-1-enyl)-2,2’-bithienyl (BBT) and T-5. The cytotoxicity was evaluated using the MTT assay, with 72 hours of incubation time. The thiophenes were identified as 5-(4-hydroxy-1-butynyl) 2,2’bityenyl (BBTOH), 5-(4-acetoxy-1-butynyl)-2-2’-bityenil (BBTOAc) and 2,2’:5’,2”-terthiophene (α-T). BBTOH, BBTAOc and α-T were active and statistically similar against the ovarian cancer cell line (OVCAR8) and the TF was statistically less active. Regarding the colon cancer cell line (HCT-116) and the central nervous system cancer, all the drugs tested were shown to be cytotoxic and statistically similar. The standard doxorubicin was shown to be statistically more active when compared to the compounds tested. These results are very impressive, since they are below the value expected by the National Cancer Institute of the United States for good antineoplastic drugs. The thiophenes caused hemolysis only in the concentrations above 250 Og/mL. Regarding the quantification through the plant development ,in general, there is no linearity about the thiophene concentration during the development of Tagetes Patula, however, can be concluded that the higher average thiophene concentration is reached in the day 60 and from this day the thiophenes concentration deacrease. Regarding the differents plant parts, the part with higher thiophene concentration is the Roots, followed by stem, flowers and stem. Nevertheless, could be observed that the maximum concentration peak is around the day 60, where the flowering period begin and then the concentration decreases. Thiophenes from T. erecta showed a good activity against cancer cell lines and a low cytotoxicity against the erythrocytes. In general, all the thiophenes isolated exhibited cytotoxic activities considered significant according to the NCI, which shows that T. erecta thiophenes represent an important class of antineoplastic inhibitors.
