CARACTERIZAÇÃO E POTENCIAL BIOTECNOLÓGICO DO PÓLEN COLETADO POR Melipona (Michmelia) seminigra seminigra NO AMAZONAS, BRASIL

Pólen apícola é um aglomerado de diversos grãos de pólen, transportados por abelhas operárias das flores de diversas espécies de plantas para seus ninhos, onde é utilizado como fonte nutritiva. O objetivo do presente trabalho foi caracterizar a microbiota do pólen fresco e desidratado coletados de colmeias de Melipona (Michmelia) seminigra seminigra, nos períodos, chuvoso (PCH) e seco (PSE), de uma comunidade situada no Baixo Amazonas, Amazonas, Brasil, no ano de 2009. Avaliar a atividade antitumoral, antimicrobiana, toxicidade e citotoxicidade dos extratos hexânico (EHEX), acetato de etila (AcOEt) e metanol (MeOH); assim como, verificar a composição físico-química, minerais e aminoácidos, além de avaliar o potencial de atividade antioxidante dos extratos MeOH, acetona 80% e hexano/diclorometano (HEX/DCM), e também os teores de vitaminas hidrossolúveis (B1, B2, B6 e C), lipossolúveis (A, E) e ß-caroteno do pólen. A ocorrência de coliformes totais, bactérias termotolerantes, fungos filamentosos e levedura foi avaliada utilizando-se diluições dos grãos de pólen em meio seletivo. Os extratos EHEX, AcOEt e MeOH foram testados na concentração de 5 µg/mL para atividade antitumoral, 5 mg/mL para citotoxicidade em eritrócitos e toxicidade frente à Artemia salina, além de 20 mg/mL para atividade antimicrobiana. Os polifenóis totais e o sequestro de radical livre foram realizados pelos métodos de Folin-Ciocalteu e DPPH (2,2-difenil-1-picrilhidrazila), tendo-se avaliados os extratos nas frações (MeOH, acetona 80% e HEX/DCM) pelo método capacidade de absorção de radicais oxigênio - ORAC. As vitaminas hidrossolúveis, lipossolúveis e ß-caroteno foram quantificadas por cromatografia líquida de alta eficiência - CLAE. A composição físico-química (proteínas, lipídeos, carboidratos, fibras e cinzas) foi realizada pela metodologia da association of official analytical chemists - AOAC. Os minerais foram detectados através de espectrofotômetro de absorção atômica e a leitura dos aminoácidos efetuada por CLAE em fase reversa a 254nm. As amostras de pólen desidratado do PCH e do PSE não apresentaram contaminação por bactérias nem fungos, tendo-se isolados 27 colônias de fungos na amostra de pólen de PCH e 26 no de PSE. Dentre estas o Penicillium spp. foi o mais frequente [PCH (96,98%) e PSE (47,7%)]. Nas condições experimentais não foi detectada atividade antitumoral dos extratos frente às linhagens tumorais MDA-MB435 (mama) e SF-295 (glioblastoma), HL-60 (leucemia) e HCT-8 (cólon) humanos. Os extratos AcOEt- PCH/PSE e MeOH-PCH demonstraram baixa toxicidade (CL50 entre 500 e 1000 µg/mL), enquanto EHEX-PCH/PSE e MeOH/PSE não foram tóxicos frente a Artemia salina (CL50>1000 µg/mL). Os extratos EHEX-PCH/PSE e AcOEt-PCH/PSE evidenciaram baixa atividade hemolítica, e inativa para o MeOH. Os extratos EHEX, AcOEt e MeOH a 20 mg/mL impediram o crescimento de Mycobacterium smegmatis, Streptoccocus mutans, Streptoccocus oralis e Candida albicans, tendo-se apresentado inativo para Escherichia coli e Staphyiloccocus aureus. O teor de fenóis totais foi de baixa atividade, mas, por outro lado, a capacidade sequestrante de radical livre por DPPH foi mais eficaz na fração acetona 80%-PSE IC50 2,53 µg/mL, e nas frações MeOH e Acetona 80%-PCH IC50 (2,74 e 2,82 µg/mL), respectivamente. Os valores da capacidade antioxidante total foi 403,45 µM TE*/g (equivalente trolox) no pólen do PCH. Das vitaminas do complexo B, apenas a B2 (riboflavina) demonstrou concentração significante, 13,4 µg/g no pólen do PSE. A somatória dos quatro homólogos de tocoferol (α+β+γ+δ) vitamina E foi (44,8 µg/g PCH e 92,0 µg/g PSE). A concentração de ß-caroteno foi 285 µg/g e vitamina A 153 UI/100g no pólen de PSE. Os teores de proteínas foram (35,99%-36,37%), lipídeos (22,77%-29,66%)
e cinzas totais (4,46%-2,96%), e os minerais Cu (70,23-183,53 µg/g), Fe (556,47- 11,1 µg/g), Ca (1,51%-1,64%) e fosfato (5,78%-7,09%), além dos aminoácidos essenciais e não essenciais. Os dados obtidos permitiram elaborar um novo produto em forma de barras nutritivas preparadas com pólen de abelhas, banana pacovi da Amazônia e castanha-do-Brasil, responsáveis pelo fornecimento de aminoácidos, ácidos graxos, carboidratos e fibras, além dos minerais: potássio, magnésio, fósforo, ferro, manganês e zinco, todos de origem vegetal, com baixo custo e potencial nutritivo para combate à desnutrição de adolescentes e adultos na faixa escolar.
Bee pollen is a cluster of many grains of pollen, transported by worker bees from the flowers of various species of plants to their nests, which is used as a nutrient source for the hive itself. The objective of this study was to characterize the microbiota of dehydrated and non-dehydrated pollen collected by Melipona (Michmelia) seminigra, in the rainy (PCH) and dry (PSE) periods, of a community located in the lower Amazon, Amazon, Brazil, in the year 2009. The aim was to evaluate the antitumor activity, antimicrobial, cytotoxicity and toxicity of the hexane extracts (EHEX), ethyl acetate (EtOAc) and methanol (MeOH); as well as, to verify the physical and chemical composition, the minerals and amino acids, and additionally to evaluate the potential antioxidant activity of the methanolic extract, 80% acetone and hexane/dichloromethane (HEX/DCM), the levels of water-soluble vitamins (B1, B2, B6 and C), liposolubles (A, E) and ß-carotene of pollen. The presence of total coliforms, thermotolerant bacteria, filamentous fungi and yeasts were assessed using dilutions of pollen grains in selective medium. The EHEX, EtOAc and MeOH extracts were tested at a concentration of 5 µg/mL for antitumor activity, 5 mg/mL for cytotoxicity in erythrocytes and toxicity against Artemia salina, and 20 mg/mL for antimicrobial activity. The total polyphenols and free radical sequestering was conducted by the methods of Folin-Ciocalteu and DPPH (2,2-diphenyl-1-picrylhydrazyl), with the extracts in the fractions (MeOH, 80% acetone and HEX/DCM) analyzed using the method of oxygen radical absorbance capacity - ORAC. Water-soluble vitamins, liposolubles and ß-carotenes were quantified by high performance liquid chromatography - HPLC. The physico-chemical composition (protein, lipid, carbohydrate, fiber and ash) was performed by the methodology of the association of official analytical chemists - AOAC. The minerals were detected by atomic absorption spectrophotometer and the reading of amino acids done by reversed-phase HPLC at 254nm. The dehydrated pollen samples of PCH and PSE did not show contamination by bacteria, with only 27 colonies of fungi identified in the fresh pollen of PCH and 26 in PSE. Among these Penicillium spp. was the most common [PCH (96.98%) and PSE (47.7%)]. Under experimental conditions antitumor activity was not detected for the extracts against the human tumor cell lines MDA-MB435 (breast) and SF-295 (glioblastoma), HL-60 (leukemia) and HCT-8 (colon). The EtOAc-PCH/PSE and MeOH-PCH extracts demonstrated low toxicity (CL50 between 500 and 1000 µg/mL), while EHEX-PCH/PSE and MeOH/PSE were not toxic against Artemia salina (CL50>1000 µg/mL). The EHEX-PCH/PSE and EtOAc-PCH/PSE extracts demonstrated low hemolytic activity, being inactive for MeOH. The EHEX, EtOAc and MeOH extracts at a concentration of 20 mg/mL inhibited the growth of Mycobacterium smegmatis, Streptococcus mutans, Streptococcus oralis and Candida albicans, and were inactive for Escherichia coli and Staphyiloccocus aureus. The total phenol content had low activity, but, on the other hand, the ability of free radical scavenging by DPPH was more effective in the 80% acetone fraction-PSE IC50 2.53 µg/mL, and in the MeOH and 80% acetone fraction-PCH IC50 (2.74 and 2.82 µg/mL), respectively. The total antioxidant capacity values were 403.45 µM TE*/g (trolox equivalent) in the pollen of PCH. Of the vitamin B complex, only B2 (riboflavin) demonstrated a concentration at 13.4 µg/g in the pollen of PSE. The sum of the four vitamin E tocopherol homologs (α+β+γ+δ) was 44.8 µg/g PCH and 92.0 µg/g PSE. The concentration of ß-carotene was 285 µg/g and vitamin A 153 UI/100g in pollen for PSE. The protein content was (35.99%-36.37%), lipids (22.77%- 29.66%), and total ash (4.46%-2.96%), and minerals, Cu had (70.23-183.53 µg/g),
Fe (556.47-811.1 µg/g), Ca (1.51%-1.64%) and phosphate (5.78%-7.09%), besides the essential and nonessential amino acids for PCH and PSE respectively. The data obtained permitted the development of a new product in the form of nutritional bars prepared with bee pollen, banana pacovi from the Amazon and Brazil nuts, which are responsible for supplying amino acids, fatty acids, carbohydrates and fiber, in addition to the minerals: potassium, magnesium, phosphorus, iron, manganese and zinc, all coming from plants, at a low cost and with the nutritional potential for combating malnutrition in adolescents and adults at the school level .
