NEUROADAPTAÇÃO INDUZIDA PELO EXERCÍCIO AERÓBICO EM CAMUNDONGOS DIABÉTICOS: ALTERAÇÕES COMPORTAMENTAIS E NEUROQUÍMICAS EM ÁREAS

Diabetes Mellitus é uma doença metabólica crônica caracterizada principalmente por episódios recorrentes de hiperglicemia devido a resistência celular à insulina ou a falta de sua produção. As complicações do diabetes atingem vários sistemas do corpo humano, como, por exemplo, o Sistema Nervoso Central (SNC). Conhecida por encefalopatia diabética, esta complicação prejudica as capacidades cognitivas de seus pacientes pela variedade de alterações fisiológicas, mais comumente, o rompimento da barreira hematoencefálica, a redução de neurotrofinas e produção excessiva de espécies reativas de oxigênio e nitrogênio. Em trabalhos prévios foram demonstrados efeitos benéficos do exercício aeróbico sobre o perfil antioxidante em diferentes doenças crónicas. Além disso, o exercício promove o aumento de fatores neurotróficos no SNC, sendo uma ferramenta importante para a melhora da memória. Assim, o objetivo deste estudo foi investigar os efeitos do exercício aeróbio crônico sobre o SNC de ratos diabéticos e suas implicações neuroquímicos e moleculares. Foram utilizados 72 camundongos fêmeas Swiss, pesando 25-30 g com água e comida ad libitum. Os animais foram inicialmente testados no teste de Open Field (CA) e em seguida no labirinto aquático de Morris (LAM) com o objetivo de dividir os animais em quatro grupos: Controle Sedentário (CS); Controle Treinado (CT); Diabéticos Sedentários (DS); e Diabetico Treinado (DT). Em seguida, os grupos DS e DT receberam 140 mg/kg de estreptozotocina, via intraperitoneal (i.p.), para indução do diabetes. Três semanas após, a glicemia foi aferida nestes grupos para confirmação do quadro hiperglicêmico, sendo assim refeito os testes comportamentais (CA e LAM) para identificação da encefalopatia diabética nos grupos diabéticos. Após os testes, os animais dos grupos CT e DT começam a executar o treinamento aeróbio através do protocolo de natação. Vinte e quatro horas após a última sessão de treinamento, os animais foram novamente testados no CA e LAM e vinte e quatro horas após a execução do teste LAM, os animais foram sacrificados e tiveram quatro áreas cerebrais retiradas: Córtex Total, Córtex Pré-frontal, Córtex Temporal, Hipocampo e Corpo Estriado. Foram aferidos peso, atividade da superóxido desmutase pela auto-oxidação da adrenalia, atividade da catalase pela degradação de peróxido de Hidrogênio, peroxidação lipídica (método TBARS), níveis de nitrito através da diazotização de nitrito e expressão protéica (BDNF, TrkB e uTrkB por western blot). Os resultados mostram que o exercício influenciou positivamente a atividade locomotora e a memória espacial em ratos diabéticos. Além disso, verificou-se melhoria do perfil antioxidante do grupo DT quando comparado com o grupo DS no hipocampo, no córtex temporal e frontal. Além disso, o exercício melhorou o estado neurotrófico do grupo DT no hipocampo. Conclui-se que o exercício promoveu efeitos benéficos em relação à cognição de ratos diabéticos provavelmente através do perfil antioxidante e neurotrófico. Diabetes Mellitus is a chronic metabolic disorder mainly characterized by recurrent episodes of hyperglycemia due the celular resistance to insulin or the lack of its production. The complications of diabetes extent for various systems along the human body such as the Central Nervous System (CNS). Also known as Diabetic Encephalopathy, this pathology impairs the cogntive capabilities of its patients by variety of symptons, being mostly commom the disrpution of the Blood Brain Barrier, the reduction of Neurotrophins and the excessive production of Reactive Oxygen and Nitrogen Especies. It has been previously shown the benefic effects of the aerobic exercise on the antioxidant profile in other chronic disorders. As well, the exercise promote the augmentation of neurotrophic factors in the CNS, being one of the most important tools for memory. Thus, the aim of this study was to investigate the effects of aerobic exercise on the CNS of diabetic rats and its neurochemical and molecular implications. It was used 72 female Swiss rats weighting 25-30g with water and food ad libitum. The animals were initially tested on the Open Field test (CA) and in Morris Water Maze test (LAM) to divide the animals in four groups: Control Sedentary (CS); Control Trained (CT); Diabetic Sedentary (DS); Diabetic Trained (DT). Then, the DS and DT group were injected with 140mg/kg of streptozotocin. Three weeks after the confirmation of the diabetes, the animals were tested again in CA and in LAM for confirmation of the diabetic encephalopathy. After these tests, the animals start to execute an aerobic training through the swimming protocol. Twenty four hours after the last training sessions, the animals were retested in LAM and CA. Twenty four hours after the execution of LAM test, the animals were euthanasied and its Total Córtex, PreFrontal Cortex, Temporal Cortex, Hippocamppus and Striatum were removed. The Catalase, Superoxide Dismutase, Malondiadehyde levels, Nitrite Levels were measured in PreFrontal Cortex, Temporal Cortex, Hippocamppus and Striatum. The protein expression of BDNF, TrkB and Trkb-T1 were measured in Hippocamppus and in Total Cortex. The results show that the exercise positivily influencied the locomotor activity and the spatial memory of diabetic rats. Also, it has been found improvements in antioxidant profile of DT group when compared to DS group in hippocamppus, Temporal and Frontal Cortex. Also the exercise improved the neurotrophic status of DT group in Hippocamppus. In sumary, the exercise exerted benefic effects regarding the cognition of diabetic rats through the antioxidant and neurotrophic profile. Keywords: Diabetes, Physical Exercise, Neurogenesis
