COMPOSIÇÃO QUÍMICA E AVALIAÇÃO DAS ATIVIDADES BIOLÓGICAS DO ÓLEO ESSENCIAL DE Vitex gardneriana SCHAUER (LAMIACEAE)

Os óleos essenciais (OEs) são produtos do metabólito secundários das plantas que possuem diversas atividades biológicas, tais como: antifúngica, anti-inflamatória, antisséptico, anti-hipertensivo, ansiolítico, anti-leishmania, antiviral, sendo também largamente utilizados na indústria alimentar, cosmética e farmacêutica. Este trabalho teve como objetivo realizar um estudo comparativo (circadiano e sazonal) do óleo essencial das folhas de V. gardneriana (OEFVG) e verificar o potencial em ensaios biológicos (anti-acetilcolinesterase; toxicidade; atividade larvicida frente ao A. aegypti e atividade antifúngica do óleo essencial. Os OEs foram extraídos através de hidrodestilação e analisados por cromatografia gasosa acoplado a espectrometria de massa (CG/EM) e cromatografia gasosa com detector por ionização de chama (CG/DIC); a avaliação da atividade inibidora da enzima aceticolinesterase foi efetuada de acordo com o ensaio enzimático e adaptada para cromatografia em camada delgada e quantitativamente realizado em placas de 96 poços de fundo chato utilizando leitor Elisa BIOTEK e a atividade antifúngica utilizando o método de microdiluição em caldo, a toxicidade foi determinada in vivo em Artemia salina e in vitro utilizando células sanguíneas. A análise do óleo essencial de V. gardneriana possibilitou a identificação de 32 componentes; os constituintes majoritários foram os sesquiterpenos cis-calameneno (23,75%) e 6,9-guaiadieno (15,12%). Na análise do ritmo circadiano foram identificados 25 constituintes (96,35%) no óleo essencial às 8h, sendo 2 monoterpenos (0,42%) e 23 sesquiterpenos (95,93%). No óleo extraido às 12h foram identificados 26 constituintes (92,26%), sendo 2 monoterpenos (0,28%) e 24 sesquiterpenos (91,98%). No óleo extraido às 17h, 32 constituintes (91,55%) foram identificados, sendo 5 monoterpenos (2,91%) e 27 sesquiterpenos (88,64%). Os principais constituintes extraidos às 8 e 12 e 17h foram os sesquiterpenos cis-calameneno (27,93, 24,48 e 25,79%), 6,9-guaiadieno (21,80, 11,19 e 18,37%) e óxido de cariofileno (16,68, 26,33 e 13,87%), respectivamente. Com relação à atividade antiacetilcolinesterásica, o OEFVG apresentou halo de inibição de 0,9 cm com valor de CI50 de 19,85 μg/mL; no teste quantitativo os óleos essenciais extraidos às 8 e 12 e 17h apresentaram CI50 de 13,04, 13,69, 11,21 μg/mL respectivamente, demonstrando um alto potencial de inibição, quando comparado ao padrão fisostigmina. No teste de toxicidade frente A. salina, o OEFVG apresentou CL50 de 98,1 μg/mL indicando uma toxicidade moderada. Já no teste de atividade hemolítica, não houve atividade nas concentrações testadas, obtendo um CI50 2,5 mg/mL. O teste larvicida contra A. aegypti apresentou CL50 de 28 g/mL considerado-o altamente ativo. Os óleos essenciais extraidos às 8 e 12 e 17h apresentaram atividade antifúngica contra estirpes de T. rubrum. Assim, os OEs de V. gardineriana mostraram serem promissores na inibição da enzima acetilcolinesterase, apresentou em excelente potencial larvicida contra o A. aegypti e atividade antifúngica.

Abstract:
The essential oils (EOS) are products of the secondary metabolite of plants that have different biological activities, such as: anticéptica antifungal, anti-inflammatory, anti-hypertensive, anxyolitic, anti-leishmania, antiviral, and also widely used in the food industry, cosmetics, pharmaceutical industry. The objective of this work was to conduct a comparative study (circadian rhythm and seasonal) of essential oil from the leaves of V. gardneriana (OEFVG) and check the potential biological assay (anti-acetylcholinesterase; tocixidade; larvicidal activity outside the A. aegypti and fungicidal activity of the essential oil. The EOs were extracted by hydrodistillation and analyzed by gas chromatography coupled to mass spectrometry (GC/MS) and gas chromatography with flame ionization detector (GC/FID); the evaluation of the inhibitory activity of the enzyme aceticolinesterase was performed according to the enzymatic assay and adapted for thin-layer chromatography and quantitatively performed on plates of 96 flat-bottomed wells using Elisa reader BIOTEK and antifungal activity using the broth microdilution method, the toxicity was determined in Artemia salina in vivo and in vitro using blood cells. The analysis of the essential oil of V. gardneriana enabled the identification of 32 constituent components; the majority was the sesquiterpenes cis-calameneno (23.75%) and 6.9-guaiadieno (15.12%). In the analysis of the circadian rhythm were identified 25 constituents (96.35%) in the essential oil at 8:00, being 2 (0.42%), monoterpenes and sesquiterpenes 23 (95.93%). The oil extracted at 12 h was identified 26 constituents (92.26%), being 2 (0.28%), monoterpenes and sesquiterpenes 24 (91.98%). The oil extracted at 17h, 32 constituents (91.55%) were identified, being 5 (2.91%), monoterpenes and sesquiterpenes 27 (88.64%). The principal constituents extracted at 8 and 12 and 17h were the sesquiterpenes cis-calameneno (27.93, 24.48 and 25.79%), 6.9-guaiadieno (21.80, 11.19 and 18.37%), caryophyllene oxide (16.68, 26.33 and 13.87%, respectively). With respect to antiacetilcolinesterásica activity, the OEFVG presented inhibition halo of 0.9 cm with IC50 value of 19.85 μg/mL; the quantitative test essential oils extracted at 8 and 12 and 17h showed IC50 of 13.04, 13.69, 11.21 μg/mL respectively, showing a high potential of inhibition, when compared to the standard physostigmine. In the toxicity test forwards A. salina, the OEFVG showed LC50 of 98.1 μg/mL indicating a moderate toxicity. Already in the hemolytic activity test, there was no activity concentrations tested, getting an IC50 2.5 mg/mL. The larvicidal test against A. aegypti showed LC50 of 28 μg/mL considered it highly active. The essential oils extracted at 8 and 12 and 17h showed antifungal activity against strains of T. rubrum. Thus, the EOS of V. gardneriana showed to be promising in the inhibition of the enzyme acetilcolonesterase, presented in excellent larvicidal potential against A. aegypti and antifungal activity.
