Bioprospecção de plantas com potencial farmacológico para o tratamento das leishmanioses

Resumo:
As leishmanioses são doenças causadas por protozoários do gênero
Leishmania que acometem milhões de pessoas em todo o mundo. Apresentam
manifestações clínicas que variam de lesões cutâneas, mucocutâneas a forma
visceral, geralmente fatal. Antimoniais pentavalentes são as drogas de primeira
escolha para o tratamento das leishmanioses, porém são tóxicas e têm
apresentado falha no tratamento devido a problemas de resistência dos
parasitas. Muita atenção tem sido dispensada a utilização de plantas como
fonte de novos medicamentos para o controle dessa doença. Nesse contexto, o
Brasil, por ser um país detentor de grande biodiversidade, possui vasto acervo
a ser explorado. O potencial farmacológico de 13 espécies vegetais de
ocorrência no Estado do Piauí para o tratamento das leishmanioses foi avaliado
por meio da atividade in vitro contra promastigotas de Leishmania amazonensis
e citotoxicidade para células de mamíferos. As espécies mais ativas foram
selecionadas para fracionamento e continuidade do estudo. Para Azadirachta
indica realizou-se fracionamento dos extratos guiado pela ação em
promastigotas para seleção das amostras mais eficientes para os testes sobre
formas amastigotas. Quatro das espécies estudadas apresentaram
considerável atividade antileishmania e baixa citotoxicidade: Protium
heptaphyllum (resina, CI50 = 5,1 µg/mL), Platonia insignis (extrato etanólico da
casca do fruto, CI50 = 22,4 µg/mL) e Zanthoxylum rhoifolium (fração hexânica
da folha, CI50 = 7,9 µg/mL). Os resultados de CI50 obtidos com o fracionamento
de A. indica para o extrato etanólico e fração diclorometano do tegumento da
amêndoa são: 2,74 e 2,08 µg/mL para promastigotas e 0,43 e 0,6 µg/mL para
amastigotas. Para o extrato etanólico e frações diclorometano e clorofórmica
obtidas das folhas foram de 38, 3,85 e 1,24 µg/mL para promastigotas e de
9,75; 1,1 e 0,55 µg/mL para amastigotas, respectivamente, em 72 horas. A
avaliação citotóxica apresentou maior seletividade para o parasita. As espécies
A. indica, P. heptaphyllum, P. insignis e Z. rhoifolium apresentaram excelente
atividade antileishmania e baixa citotoxicidade. As atividades das plantas
associadas à segurança apresentada estimulam a continuidade da pesquisa na
busca de substâncias isoladas e posterior desenvolvimento de formulações

Abstract:
Leishmaniasis are diseases caused by protozoa of the genus Leishmania that
affect millions of people around the world. They vary in clinical manifestations
ranging from skin lesions, mucocutaneous to the visceral form, usually fatal.
Pentavalent antimonials are the drugs of first choice for the treatment of
leishmaniasis, but are toxic and have shown treatment failure due to problems
of parasite resistance. Significant attention has been given to the use of plants
as source of new medicines to the control of this disease. In this context, Brazil,
a country with huge biodiversity, has vast acquis to be explored. The
pharmacological potential of 13 plant species occurring in the State of Piauí for
the treatment of leishmaniasis was evaluated through in vitro activity against
promastigote of Leishmania amazonensis and safety for mammalian cells. The

For Azadirachta indica a fractionation of the extracts was performed by the
action on promastigotes to the selection of the more efficient samples for tests
on amastigotes forms. Four of the species studied showed considerable
antileishmania activity and low cytotoxicity: Protium heptaphyllum (resin, IC50 =
5.1 µg/mL), Platonia insignis (ethanolic extract from the fruit, IC50 = 22.4 µg/mL)
and Zanthoxylum rhoifolium (hexane fraction from leaves, IC50 = 7.9 µg/mL).
The results of the IC50 obtained with the fractionation of A.indica for the ethanol
extract and dichloromethane fraction of the almond tegument are: 2.74 and 2.08
µg/mL for promastigotes and 0.43 and 0.6 µg/mL for amastigotes. To the
dichloromethane and chloroform fractions obtained from the ethanol extract of
leaves was 38, 3.85 and 1.24 µg/mL for promastigotes and 9.75, 1.1 and 0.55
µg/mL for amastigotes, respectively, in 72 hours. The cytotoxic evaluation
showed higher selectivity for the parasite. The species A. indica, P.
heptaphyllum, P. insignis and Z. rhoifolium showed excellent antileishmania
activity and low cytotoxicity. The activities of plants associated to the safety
shown stimulate the continuation of research in the pursuit of isolated
substances and further development of formulations for the treatment of
