Resumo:
O Croton argyrophylloides Muell. Arg. é conhecido popularmente no Nordeste como
marmeleiro prateado. Dentre os constituintes presentes no óleo essencial do C.
argyrophylloides (OECa) temos β-cariofileno (β-CAR) que é um terpenóides com
uma atividade biológica bastante significativa. O objetivo desse trabalho foi verificar
o efeito do OECa e de um dos seus constituintes o β-CAR em músculo liso da veia
porta, íleo e bexiga. Foi observado que o OECa e o β-CAR possui efeito tanto no
acoplamento eletromecânico como no farmacomecânico. No acoplamento
eletromecânico a EC50 na presença do OECa para a contração fásica foi de 19,21 ±
1,65 µg/ml, 104,27 ± 8,19 µg/ml e 343,54 ± 11,71µg/ml respectivamente para veia
porta, íleo e bexiga. Na contração tônica, a EC50 para veia porta, íleo e bexiga foi de
7,81 ± 2,02 µg/ml, 29,99 ± 8,44 µg/ml e 92,50 ± 11,00 µg/ml. Com o β-CAR, a EC50
da fase fásica foi de 92,64 ± 5,34 µg/ml, 15,57 ± 6,38µg/ml e 214,93 ± 11,06 µg/ml
respectivamente para veia porta, íleo e bexiga. A fase tônica na presença do β-CAR
teve como EC50 24,76 ± 9,63 µg/ml, 12,54 ± 1,43 µg/ml e 181,19 ± 6,66 µg/ml
respectivamente para veia porta, íleo e bexiga. No acoplamento farmacomecânico a
EC50 na presença do OECa foi de 38,53 ± 14,83 µg/ml, 125,93 ± 10,91 µg/ml e
122,19 ± 57,33 µg/ml respectivamente para veia porta, íleo e bexiga, na contração
fásica. Na contração tônica a EC50 foi de 16,81 ± 2,70 µg/ml, 47,97 ± 08,09 µg/ml e
115,71 ± 20,51 µg/ml, respectivamente em veia porta, íleo e bexiga. Em relação ao
β-CAR, esse efeito foi um pouco menos expressivo nas duas fases da contração,
entre os três tecidos. Com β-CAR a EC50 foi de 101,59 ± 2,81 µg/ml, 86,73 ± 12,12
µg/ml e 560,20 ± 61,88 µg/ml, respectivamente em veia porta, íleo e bexiga, na fase
fásica e de 41,45 ± 12,89 µg/ml, 105,09 ± 4,09 µg/ml e 461,06 ± 7,13 µg/ml,
respectivamente em veia porta, íleo e bexiga, na fase tônica. Em relação a contração
induzida por CaCl2 e BaCl2, também foi observado efeito tanto do OECa quanto do
β-CAR nos três tecidos. Os resultados sugerem que tanto o OECa como o β-CAR
possuem efeito nos três tecidos, mas com uma certa especificidade pela veia porta.
Foi observado que tanto o OECa como o β-CAR possuem um maior efeito no
acoplamento eletromecânico que no farmacomecânico.O estudo sugere que o efeito
antiespasmódico do Croton argyrophylloides e β-cariofileno no músculo liso de íleo,
veia porta e bexiga, pode ter ocorrido pelo bloqueio dos canais de cálcio, sendo
predominante esse efeito nos canais do tipo VOC.

Abstract:
Croton argyrophylloides Muell. Arg. is popularly known in Northeast like quince
silver. Among the constituents of the essential oil of C. argyrophylloides (OECa) have
β-caryophyllene (β-CAR) which is a terpenoid with biological activity very
significantly. The aim of this study was to assess the effect of OECa and one of its
constituent β-CAR in the smooth muscle of the portal vein, ileum and bladder. It was
observed that OECa and β-CAR has effect on electromechanical coupling and in
pharmacomechanical. Electromechanical coupling in the EC50 in the presence of
OECa for phasic contraction was 19.21 ± 1.65 µg/ml, 104.27 ± 8.19 µg/ml and
343.54 ± 11.71 µg/ml respectively for the portal vein, ileum and bladder. In the tonic
contraction, the EC50 for the portal vein, ileum and bladder was 7.81 ± 2.02 µg/ml,
29.99 ± 8.44 µg/ml and 92.50 ± 11.00 mg µg/ml. Regarding β-CAR, the EC50 of
phasic phase was 92.64 ± 5.34 µg/ml, 15.57 ± 6.38 µg/ml and 214.93 ± 11.06 µg/ml
respectively for the portal vein, ileum and bladder. The tonic phase in the presence of
β-CAR EC50 was 24.76 ± 9.63 µg/ml, 12.54 ± 1.43 µg/ml and 181.19 ± 6.66 µg/ml
respectively for the portal vein, ileum and bladder. Coupling pharmacomechanical the
EC50 in the presence of OECa was 38.53 ± 14.83 µg/ml, 125.93 ± 10.91 µg/ml and
122.19 ± 57.33 µg/ml respectively for the portal vein, ileum and bladder phasic
contraction. Tonic contraction in the EC50 was 16.81 ± 2.70 µg/ml, 47.97 ± 08.09
µg/ml and 115.71 ± 20.51 µg/ml, respectively in the portal vein, ileum and bladder. βCAR had less effect in both the expressive phase of contraction among the three
tissues.B-CAR with the EC50 was 101,59 ± 2,81 µg/ml, 86,73 ± 12,12 µg/ml and
560,20 ± µg/ml, respectively in the portal vein, ileum and bladder during phasic and
41.45 ± 12.89 µg/ml, 105.09 ± 4.09 µg/ml and 461.06 ± 7.13 µg/ml, respectively in
the portal vein, ileum and urinary bladder in the tonic phase . For contraction induced
by CaCl2 and BaCl2 effect was also observed both OECa as the β-CAR in the three
tissues. The results suggest that both the OECa as β-CAR have effect in all three
tissues, but with some specificity the portal vein. It was observed that both the OECa
as β-CAR have a greater effect on electromechanical coupling than
pharmacomechanical .O study suggests that the antispasmodic effect of Croton
argyrophylloides and β-caryophyllene in the smooth muscle of the ileum, portal vein
and bladder, may have occurred by blocking calcium channels, with predominant
effect on this type VOC channels.
